1 About This Book
The GxP Statistical Computing Environment is a review and practitioner’s guide to building regulated clinical pipelines and the products around them — what the field knows, what works in production, and what is still missing.
It answers, in one place, three questions practitioners keep asking:
- What does the field actually require? A grounded tour of the regulatory frame (risk-based, evidence-driven, deliberately non-prescriptive), the validation literature from screening to intended-use evidence, the platform landscape from validated monoliths to open ecosystems, and the pipeline and standards machinery (locks, graphs, ARD, Dataset-JSON).
- How do you build one? A seven-layer reference architecture for a trust pipeline — catalog, experience, identity, orchestration, execution, evidence, data — plus two in-production case studies: a twenty-one-service composable review platform and a validation gateway with metamorphic testing and governed LLM assistance.
- What should be true of any such system? Nine principles — spec in/evidence out, contracts as constitution, artifact addressing, independent verification that stops the line, validation as deliverable, honesty by design, red lines and the human in the loop, absorb-don’t-rebuild, one product/many homes — each with a test you can run on any system, including your own.

1.2 How it is organized
Part I fixes definitions and states the thesis. Part II is the literature review, with references collected in a dedicated chapter. Part III states the nine principles. Part IV builds the factory: reference architecture, cases, and the catalog as front door. Part V is the long game — open-core business, operating habits, and the field’s open gaps. Chapters are short and cross-referenced; the book can be read front to back or entered anywhere.
1.3 Sources and honesty
External claims are cited inline and listed with verification status in the References chapter; the underlying literature review is preserved in the book’s source repository. The case studies are the author’s own production systems, reported with their boundaries — including where each deliberately stops short of GxP claims. A book about evidence that overstated its own would refute itself.
